What changes are there to the minimum requirements for playing GM2 games? I think I remember reading it no longer supports Windows Vista right? And you need the new Direct X? Will this causes problems if I realease my game on itchio?

Doesn't support XP, does support Vista but now uses DirectX 11 so Vista users will need to be pointed towards the DX11 Installer if they don't already have it.
