how to convert PNG 32bit and 24bit in color depth to high color 16 bit (RGBA 5: 5: 5: 1) who tried it? Very few photographers and editors convert exactly to 16-bit with an alpha channel, please tell me who knows?

Are you asking how to do this in code, or asking about software that can perform such conversion tasks? You posted this in the Programming forum, hence the question.

GIMP will export to the more niche PNG pixel formats.

Just to make sure, are we talking about exporting color PNGs with a 1-bit alpha/mask? I didn't even know this was possible with PNG.

RGBA 5:5:5:1 is not 16bit, but 32 indexed colours and 1 bit transparency.

Converting 32bit/24bit to 16 bit per channel makes no sense - it would not increase the colour range.

Perhaps explain again what your exact requirement would be for those images?
